NY-ESO-1 (Cancer/Testis Antigen 1B) is a protein typically expressed only in the testis and placental cells but aberrantly overexpressed in numerous cancers, making it an ideal immunotherapy target (UniProt P78358). The specific peptide sequence SLLMWITQC (amino acids 157-165) is processed and presented on the cell surface by HLA-A*02:01 or HLA-A*02:06 molecules (PMID: 10944120). This peptide-MHC complex is the primary recognition site for several high-affinity T-cell receptor (TCR) therapies, such as afamitresgene autoleucel, which was FDA-approved in 2024 for synovial sarcoma (FDA.gov). By engineering a patient's T-cells to express a TCR specific to this complex, the immune system can selectively identify and eliminate malignant cells while sparing most healthy tissues. This target has shown significant clinical efficacy in treating solid tumors such as synovial sarcoma and myxoid/round cell liposarcoma (Adaptimmune.com).
