NY-ESO-1 (157-165) peptide presented by Human Leukocyte Antigen A*02:03, A*02:05, A*02:06, and A*02:10 is a specific peptide-major histocompatibility complex (pMHC) target used in cancer immunotherapy. The peptide, with the sequence SLLMWITQC, is derived from the Cancer-Testis Antigen 1B (CTAG1B), commonly known as NY-ESO-1 (UniProt: P78358). NY-ESO-1 is highly expressed in various malignancies, including synovial sarcoma and melanoma, while its expression in normal adult tissues is restricted to the testis and placenta (PubMed: 16322273). This restricted expression pattern makes it an ideal target for reducing off-tumor toxicity in therapeutic applications. This specific pMHC complex is recognized by engineered T-cell receptors (TCRs) in therapies like afamitresgene autoleucel, which was recently approved for synovial sarcoma (FDA: Tecelra Approval 2024). By targeting these specific HLA-A*02 alleles, the therapy can be applied to a broader patient population beyond those carrying the more common HLA-A*02:01 allele (PubMed: 32661151). Upon binding, the engineered T cells are activated to release cytotoxic granules and cytokines, resulting in the targeted destruction of cancer cells. Clinical development of drugs targeting this complex has shown significant efficacy in treating advanced solid tumors.
