Overview

The NY-ESO-1 peptide 157–165 presented by HLA-A*02:01 is a peptide–MHC class I complex formed from the cancer-testis antigen NY-ESO-1, mainly expressed in germ cells but aberrantly expressed in various tumors. Its SLLMWITQC peptide sequence is displayed via HLA-A*02:01 molecules on cancer cells, triggering a strong immune response due to its high immunogenicity. Immunotherapies—such as T cell receptor–engineered T cells and peptide vaccines—target this complex to selectively attack tumor cells in patients expressing HLA-A*02:01 and NY-ESO-1. Clinical trials show promising efficacy in several cancers, with limited safety concerns due to the antigen’s tumor-restricted expression; however, immune escape through antigen loss or MHC downregulation can limit long-term responses[4][5][6][1][2][3][8].
