Overview

The NY-ESO-1 peptide-HLA-A*02:01 complex is a molecular assembly formed when the immunogenic 9-mer NY-ESO-1 peptide (most commonly SLLMWITQV, amino acids 157–165) binds in the cleft of the HLA-A*02:01 molecule, a prevalent MHC class I allele. NY-ESO-1 is a cancer-testis antigen absent from normal adult tissues except for the testis and placenta, but highly expressed in a broad range of cancers, making it an ideal tumor target. The peptide-HLA complex is recognized by cytotoxic T cells, either naturally occurring or engineered, thereby mediating immune attack against tumor cells bearing NY-ESO-1. Structural studies confirm the stability and immunodominance of this complex, and its safety and effectiveness have been demonstrated in various immunotherapy trials, including TCR-T cell therapy and peptide vaccine approaches[1][3][4][6][7].
