Orotidine 5'-phosphate decarboxylase (ODCase) is a crucial enzyme in the de novo biosynthesis of pyrimidine nucleotides. It catalyzes the decarboxylation of orotidine 5′-monophosphate (OMP) to uridine 5′-monophosphate (UMP), which is essential for RNA and DNA synthesis. ODCase accelerates the nonenzymatic rate of OMP decarboxylation by a factor of approximately 10^17, making it one of the most proficient enzymes known. It is a drug target against pathogens and also used as a selection marker in yeast strain engineering.
