Overview

PorB is the major outer membrane porin protein of Neisseria gonorrhoeae—a beta-barrel channel protein essential for the bacterium's survival and responsible for ion exchange between the organism and its environment, as well as for mediating host-pathogen interactions. PorB plays a crucial role in immune evasion by binding complement regulatory proteins, modulating apoptosis, and inhibiting the oxidative burst in polymorphonuclear leukocytes, thereby increasing bacterial persistence. OMPs are highly expressed, immunogenic surface proteins and are investigated as potential vaccine antigens and therapeutic targets; however, sequence variability and essentiality present significant challenges. OMPs also affect antibiotic susceptibility, either directly as portals for diffusion or as components of efflux systems (e.g., MtrE). Other OMPs (Opa, BAM complex) have related but distinct roles in pathogenesis, host cell invasion, and membrane structure.

Mechanism of action

Drugs may inhibit ion channel function, block antibiotic efflux, or target PorB for immune modulation (e.g., vaccine antigen candidates). Modulation of host immune response, e.g., binding complement regulatory proteins to avoid killing. Vaccine candidates try to elicit antibodies against PorB for protective immunity.
