Overview

The Ovalbumin-specific CD4+ T-cell receptor (OT-II TCR) is a specialized antigen receptor expressed on CD4+ T-cells that specifically recognizes the 323–339 peptide fragment of chicken ovalbumin presented by the MHC class II molecule I-Ab (Barnden et al., 1998, Immunology and Cell Biology). This TCR is a cornerstone of immunological research, particularly through the use of OT-II transgenic mice, which allow for the tracking of antigen-specific T-cell responses in vivo (Robertson et al., 2000, Journal of Immunology). Upon ligation with its cognate peptide-MHC complex, the TCR initiates a signaling cascade through the CD3 complex, activating pathways such as the NFAT and NF-κB pathways, which lead to T-cell proliferation and cytokine secretion (Jenkins et al., 2001, Annual Review of Immunology). While primarily utilized as a research model for studying T-cell differentiation and tolerance, the OT-II system is also used to evaluate the pharmacodynamics of immunosuppressive agents like calcineurin inhibitors and costimulation blockers (Murphy et al., 1990, Science). It serves as a critical tool for validating novel immunotherapies, including TCR-engineered T-cell therapies and vaccine candidates, by providing a predictable and measurable immune response.
