Overview

p21-activated kinase 1 (PAK1) mRNA is the messenger RNA transcript that encodes the PAK1 protein, a member of the Group I p21-activated kinases. PAK1 serves as a critical molecular switch, integrating signals from Rho GTPases like Rac1 and Cdc42 to regulate cytoskeletal dynamics, cell migration, and cell cycle progression (UniProt P35052). In various malignancies, including breast, ovarian, and pancreatic cancers, PAK1 mRNA is frequently overexpressed, driving oncogenic signaling pathways such as MAPK and PI3K/Akt (PubMed: 29731914). Targeting the mRNA transcript via RNA interference (RNAi) or antisense oligonucleotides (ASOs) offers a therapeutic strategy to silence PAK1 expression, thereby inhibiting tumor invasion and overcoming resistance to conventional therapies (PubMed: 24603390). This approach is particularly relevant given the difficulty in developing highly selective small-molecule inhibitors for the PAK1 kinase domain. Additionally, PAK1 mRNA levels have been linked to viral entry and replication, making it a potential target for antiviral interventions (PubMed: 32454402).
