Paracrine signaling is a form of cell-to-cell communication in which a cell produces signaling molecules that affect nearby target cells within the same tissue or microenvironment. In immunomodulation, paracrine signaling plays a crucial role by mediating interactions between various immune cells and their environment. Key signaling molecules include cytokines (e.g., IL-10, TNF-α), chemokines, growth factors, prostaglandins, and indoleamine 2,3-dioxygenase (IDO). Source cells, such as mesenchymal stromal/stem cells (MSCs), secrete these factors, which then bind to receptors on target cells like T cells, B cells, NK cells, DCs, and macrophages, influencing their behavior. This pathway is involved in suppressing or enhancing inflammatory responses, promoting tissue repair, and regulating adaptive immunity. Understanding paracrine signaling has led to therapeutic strategies using MSCs or their secretome for treating autoimmune diseases or promoting regeneration after injury.
