Overview

The Paraventricular nucleus of the hypothalamus (PVN) is a critical anatomical structure located in the hypothalamus that serves as a major integration center for the endocrine and autonomic nervous systems (StatPearls, 2023). It contains magnocellular neurons that produce oxytocin and vasopressin, and parvocellular neurons that secrete corticotropin-releasing hormone (CRH), the primary initiator of the hypothalamic-pituitary-adrenal (HPA) axis (Wikipedia, 2024). The PVN plays a central role in regulating the body's response to stress, fluid balance, and cardiovascular function (Bains et al., 2015). Dysregulation of PVN activity is associated with various pathologies, including chronic hypertension, obesity, and psychiatric conditions like depression and anxiety (Ferguson et al., 2008). Although the PVN is a brain region rather than a single molecular target, it contains numerous receptors, such as the melanocortin-4 receptor (MC4R), which are targeted by drugs like setmelanotide to treat metabolic disorders (NIH, 2023).
