The Parkin mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the PRKN (formerly PARK2) gene transcript, which encodes the E3 ubiquitin-protein ligase Parkin. This region contains multiple binding sites for microRNAs (miRNAs), such as miR-103a-3p, miR-218, and miR-181a, which post-transcriptionally downregulate Parkin expression by promoting mRNA degradation or inhibiting translation. In Parkinson's disease, particularly early-onset autosomal recessive forms, Parkin levels or activity are often reduced, leading to impaired mitophagy and the accumulation of damaged mitochondria. Therapeutic strategies targeting the Parkin mRNA 3'-UTR aim to increase Parkin protein levels by using antisense oligonucleotides (ASOs) or miRNA inhibitors (antagomirs) to block these inhibitory interactions. By restoring Parkin expression, these therapies seek to enhance mitochondrial quality control and provide neuroprotection to dopaminergic neurons. Beyond neurodegeneration, the Parkin 3'-UTR is also studied in oncology, as Parkin acts as a tumor suppressor in several cancers where its expression is frequently silenced.
