Overview

PAS domain-containing protein 1 (PASD1) is a transcription factor and a member of the cancer-testis antigen (CTA) family, primarily known for its role in regulating the biological clock and its restricted expression pattern. In healthy individuals, PASD1 expression is largely confined to the testis, but it is aberrantly upregulated in various malignancies, including diffuse large B-cell lymphoma (DLBCL), acute myeloid leukemia (AML), and several solid tumors (UniProt Q8IV35; PMID: 16452186). Functionally, PASD1 acts as a negative regulator of the circadian rhythm by suppressing the transcriptional activity of the CLOCK-BMAL1 complex, a mechanism that may contribute to oncogenesis by disrupting normal cellular cycles (PMID: 25770204). Due to its high immunogenicity and tumor-restricted expression, PASD1 is an attractive target for cancer immunotherapy, particularly for the development of cancer vaccines and T-cell-based therapies (PMID: 21441915). Clinical interest in PASD1 also extends to its use as a biomarker for disease progression and patient stratification in hematological cancers. While no small molecule inhibitors are currently approved, experimental approaches focusing on peptide vaccines have shown potential in inducing specific T-cell responses against PASD1-positive tumor cells (PMID: 17634404).
