Phosphatidylinositol-4,5-bisphosphate 4-phosphatase 1 (PIP4P1) is a transmembrane enzyme that hydrolyzes phosphatidylinositol-4,5-bisphosphate (PtdIns-4,5-P2), removing the 4-phosphate to generate phosphatidylinositol-4-phosphate (PtdIns-4-P)[4]. It plays a role in cellular lipid signaling, influences membrane trafficking events, and regulates the positioning and transport of lysosomes by recruiting JIP4 to lysosomal membranes[4]. PIP4P1 is also involved in the assembly of the lysosomal V-ATPase complex and activation of the mTORC1 pathway in response to amino acids; emerging research suggests it may have a role in cholesterol metabolism and certain cancers[4]. There are no approved drugs directly targeting PIP4P1, and its use as a clinical biomarker or drug target is still exploratory.
