Phosphatidylinositol 4-phosphate 3-kinase catalytic subunit type 2 beta (PI3KC2β) is an enzyme belonging to the Class II phosphoinositide 3-kinase (PI3K) family, characterized by its unique C-terminal C2 domain (UniProt: O00750). It primarily catalyzes the phosphorylation of phosphatidylinositol (PI) to PI3P and PI4P to PI(3,4)P2, playing a vital role in clathrin-mediated endocytosis and vesicular trafficking (PubMed: 29533395). PI3KC2β is activated by various growth factor receptors, such as EGFR and PDGFR, and is involved in regulating cell migration and survival (PubMed: 11274168). In clinical contexts, its overexpression is linked to increased malignancy and drug resistance in cancers like glioblastoma and small cell lung cancer (PubMed: 21832177). While most current PI3K inhibitors target Class I isoforms, PI3KC2β is gaining attention as a potential target to disrupt endosomal signaling pathways in cancer and metabolic diseases (PubMed: 30635424).

Mechanism of action

ATP-competitive inhibition of the kinase domain to prevent the phosphorylation of phosphatidylinositol into phosphatidylinositol 3-phosphate (PI3P) and phosphatidylinositol 4-phosphate into phosphatidylinositol 3,4-bisphosphate (PI(3,4)P2).
