Overview

The PI3K-Akt signaling proteins constitute a critical intracellular pathway that translates extracellular signals from growth factors and cytokines into essential cellular responses [1.2.1, 1.4.1]. This pathway is centered around the activation of phosphoinositide 3-kinase (PI3K), which generates the second messenger phosphatidylinositol (3,4,5)-trisphosphate (PIP3), leading to the recruitment and activation of the serine/threonine kinase Akt (also known as Protein Kinase B) [1.3.1, 1.4.4]. Once active, Akt phosphorylates a multitude of downstream targets, including mTOR, GSK3, and FoxO transcription factors, to regulate cell cycle progression, metabolism, and survival [1.3.1, 1.4.3]. Dysregulation of this pathway, often through PIK3CA mutations or PTEN loss, is a hallmark of many human cancers and contributes to therapy resistance [1.3.5, 1.4.1]. Consequently, numerous small-molecule inhibitors targeting various components of the PI3K-Akt axis have been developed and approved for clinical use, particularly in oncology [1.4.2, 1.3.3]. Beyond cancer, the pathway plays a vital role in insulin signaling and metabolic homeostasis, making it a focus for research in diabetes and cardiovascular health [1.2.2, 1.4.1].
