Overview

PITPNA antisense RNA 1 (PITPNA-AS1) is a long non-coding RNA transcribed on the antisense strand of the PITPNA locus. It is predominantly found in the cytoplasm, where it fulfills its regulatory function chiefly through microRNA sponging, acting as a competing endogenous RNA (ceRNA). Elevated PITPNA-AS1 expression has been documented in several cancer types, particularly hepatocellular carcinoma and lung cancer, where it is associated with enhanced cellular proliferation, migration, invasion, and poor patient outcomes. Mechanistically, PITPNA-AS1 promotes cancer progression by binding and sequestering tumor-suppressive microRNAs (miR-448, miR-363-5p, miR-32-5p, and others), resulting in the derepression of oncogenic targets such as ROCK1, PDGFD, and HMGB3. These activities make PITPNA-AS1 both a promising therapeutic target and biomarker for cancer diagnosis, prognosis, and patient stratification.

Mechanism of action

Cancer therapy strategies target PITPNA-AS1 for silencing or knockdown to inhibit its oncogenic functions (e.g., shRNA-mediated silencing). PITPNA-AS1 acts as a sponge for tumor suppressive microRNAs, thus derepressing target mRNAs that drive tumor progression (e.g., sponging miR-448 to increase ROCK1 expression, miR-363-5p to regulate PDGFD, miR-32-5p in NSCLC, miR-876-5p in HCC).
