Overview

PIWI-interacting RNA 5939 (piRNA-5939) is a small non-coding RNA that has been identified as a functional oncogenic driver in lung cancer progression [2]. It primarily functions through the PIWIL3-dependent post-transcriptional regulation of genes involved in cell growth and migration [2, 3]. In lung cancer cells, piRNA-5939 is found in both the cytoplasm and the nucleus, where it modulates the expression of PIWIL3 mRNA and protein [2]. High expression of this piRNA promotes oncogenic signaling pathways, including those responsible for cell cycle progression and cytoskeletal remodeling [2, 4]. Conversely, the silencing of piRNA-5939 has been shown to significantly reduce cell proliferation by approximately 40% and migration by 55% in experimental models [2, 3]. This regulatory axis represents a previously unrecognized mechanism in lung cancer and suggests that piRNA-5939 is a potential therapeutic vulnerability [2, 5]. Beyond its role as a target, piRNA-5939 is also being explored as a diagnostic biomarker for non-small cell lung cancer (NSCLC) [2, 5]. Although no small-molecule drugs currently target this RNA, it is a candidate for antisense oligonucleotide-based therapeutic strategies [6]. Its specific expression in tumor tissues compared to normal somatic tissues makes it an attractive target for precision medicine [6]. The piRNA-5939/PIWIL3 axis highlights the emerging importance of the piRNA pathway in somatic cancer biology [6, 10].
