Overview

Plasmodium falciparum hexose transporter 1 (PfHT1) is a vital membrane protein that facilitates the uptake of glucose and fructose into the malaria parasite (UniProt Q9N6L3). Because the erythrocytic stages of P. falciparum are entirely dependent on the rapid consumption of host glucose for energy via glycolysis, PfHT1 serves as a critical metabolic bottleneck (Krishna et al., 2000). It is a member of the Major Facilitator Superfamily (MFS) and is expressed throughout the parasite's life cycle in the human host (Joachimiak et al., 2020). Research has shown that PfHT1 is essential for parasite survival, as its genetic knockdown or chemical inhibition leads to parasite death by starvation (PubMed: 10792020). The transporter's unique binding pocket, which can accommodate both glucose and fructose, provides a structural basis for designing selective inhibitors that do not cross-react with human glucose transporters like GLUT1 (Huang et al., 2021). Consequently, PfHT1 is a highly validated target for the development of new antimalarial drugs aimed at overcoming resistance to current therapies (Nature Communications, 2020).
