Plasmodium falciparum merozoite surface protein 2 (MSP2) is a major glycosylphosphatidylinositol (GPI)-anchored protein found on the surface of the malaria merozoite [1]. The 3D7 allele represents one of the two primary dimorphic families of MSP2, characterized by specific repetitive sequences in its central variable domain [2]. MSP2 is an intrinsically disordered protein that plays a vital role in the structural integrity of the merozoite surface and is essential for the invasion of host erythrocytes [3]. Because it is highly immunogenic and targeted by protective antibodies in naturally infected individuals, it is a prominent candidate for recombinant malaria vaccines [4]. Therapeutic strategies focusing on the 3D7 allele aim to induce strain-specific or cross-reactive antibodies that block parasite entry into red blood cells or facilitate clearance via antibody-dependent cellular inhibition [5]. The high degree of polymorphism in MSP2 serves as a mechanism for the parasite to evade the host immune system, making the inclusion of multiple allelic variants a necessity for broad vaccine efficacy [2, 5]. Clinical evaluation of MSP2-based vaccines has shown they are safe and capable of reducing parasite density in vaccinees [4].
