Overview

Platelet basic protein (CXCL7), whose major proteolytic cleavage product is Beta-thromboglobulin, is a member of the CXC chemokine family primarily stored in the alpha-granules of platelets [UniProt: P02775]. Upon platelet activation and subsequent degranulation, it is released into the vascular environment where it serves as a sensitive indicator of pro-thrombotic activity and mediates the chemoattraction and activation of neutrophils through CXCR1 and CXCR2 receptors [PubMed: 15927447]. Beyond its role in acute thrombosis, it is involved in various physiological and pathological processes including wound healing, fibroblast proliferation, and the inflammatory response in atherosclerosis [PubMed: 8316102]. Clinically, Beta-thromboglobulin is recognized as a specific biomarker for platelet-related disorders, with elevated levels frequently observed in patients with myocardial infarction, deep vein thrombosis, and diabetes-associated vascular complications [NCBI: NBK557451]. While it is not a direct therapeutic target for current pharmaceuticals, its release is modulated by anti-platelet agents such as aspirin and P2Y12 inhibitors [PubMed: 2262174].

Mechanism of action

There are no approved drugs that directly bind to or inhibit Beta-thromboglobulin; however, its systemic levels are reduced by anti-platelet therapies that inhibit platelet activation and the subsequent release of alpha-granules.
