Overview

Platelet-derived growth factor receptor beta (PDGFRβ) is a transmembrane receptor tyrosine kinase (UniProt: P09619) that plays a pivotal role in the development and maintenance of mesenchymal tissues. It is highly expressed on activated myofibroblasts and pericytes, where it mediates signals for cell proliferation, migration, and extracellular matrix production in response to PDGF ligands (PubMed: 29109500). In chronic inflammatory and fibrotic diseases, the persistent activation of PDGFRβ on myofibroblasts drives the excessive accumulation of connective tissue, leading to organ dysfunction in the lungs, liver, and kidneys (PubMed: 24613813). Additionally, PDGFRβ signaling in the tumor microenvironment supports angiogenesis and increases interstitial fluid pressure, which can hinder drug delivery (PubMed: 15548691). Therapeutic strategies targeting PDGFRβ primarily involve small-molecule tyrosine kinase inhibitors, such as nintedanib and imatinib, which block its signaling to arrest fibrotic progression and inhibit tumor growth (NIH: PubChem CID 9823820).
