Overview

Plexin-B2 (PLXNB2) is the primary functional cell surface receptor for Angiogenin (ANG), a member of the ribonuclease A superfamily and a potent inducer of neovascularization (PubMed: 29100074). The binding of Angiogenin to Plexin-B2 triggers receptor-mediated endocytosis and translocation of the ligand to the nucleus, where it stimulates ribosomal RNA transcription to drive cell growth and survival (UniProt: O15031). In the context of oncology, the ANG-PLXNB2 axis is upregulated in various solid and hematopoietic cancers, including glioblastoma and prostate cancer, where it supports tumor angiogenesis and maintains cancer stem cell populations (PubMed: 35418214). Conversely, mutations or loss of function in this signaling pathway are associated with neurodegenerative disorders such as amyotrophic lateral sclerosis (ALS), highlighting its role in neuroprotection (PubMed: 18604453). Therapeutic interventions targeting this receptor, such as monoclonal antibodies, aim to disrupt the oncogenic signaling and neovascularization promoted by this complex.

Mechanism of action

Inhibition of the angiogenin-plexin-B2 signaling axis by blocking ligand binding, preventing receptor-mediated endocytosis, or inhibiting the subsequent nuclear translocation of angiogenin to suppress ribosomal RNA transcription and tumor cell proliferation.
