Overview

PMS1 homolog 2, mismatch repair system component pseudogene 10 (PMS2P10, also called PMSR4) is a nonfunctional DNA sequence on chromosome 7 closely related to the PMS2 gene, which encodes a key DNA mismatch repair enzyme[1][3][5]. PMS2P10 is one of several PMS2 pseudogenes found in this region; it does not produce a protein or participate in mismatch repair. The presence of closely related pseudogenes such as PMS2P10 complicates genetic testing for PMS2 mutations, which are implicated in DNA mismatch repair deficiency syndromes and Lynch syndrome. However, PMS2P10 itself is not a therapeutic target, does not have biological function, and is not implicated directly in disease or therapy. Key considerations: - PMS2P10 is frequently misinterpreted as the functional PMS2 gene due to high sequence similarity and misleading nomenclature in genetic reports. - Only PMS2 (not PMS2P10) is a functional DNA mismatch repair enzyme involved in cancer predisposition and cell cycle regulation[1][3][5]. - If seeking structured data for therapeutic targets, PMS2P10 should be excluded or flagged as a pseudogene. Only the canonical PMS2 gene is relevant for functional and disease roles.
