Overview

Poliovirus receptor-related protein 2 (Nectin-2), also known as CD112, is a calcium-independent cell-cell adhesion molecule belonging to the immunoglobulin superfamily [1]. It is primarily localized at adherens junctions in epithelial and endothelial cells, where it plays a crucial role in organizing cell-cell junctions and regulating cell signaling [1, 2]. In the context of immunology, Nectin-2 serves as a ligand for several receptors on T cells and natural killer (NK) cells, including the inhibitory receptors PVRIG (CD112R) and TIGIT, as well as the costimulatory receptor DNAM-1 (CD226) [2, 3]. Overexpression of Nectin-2 is frequently observed in various malignancies, including breast, ovarian, and lung cancers, where it contributes to immune evasion by preferentially binding to inhibitory receptors over activating ones [3]. Consequently, Nectin-2 has emerged as a promising therapeutic target in immuno-oncology, with several monoclonal antibodies and combination therapies currently under investigation to block its inhibitory interactions and restore anti-tumor immunity [4].
