Poly(A) polymerase gamma (PAPOLG) is a nuclear enzyme in the poly(A) polymerase family responsible for the post-transcriptional adenylation of the 3'-end of mRNA precursors and several small RNAs, including signal recognition particle (SRP) RNA, nuclear 7SK RNA, U2 small nuclear RNA, and ribosomal 5S RNA[1][2][8]. PAPOLG catalyzes the template-independent addition of adenosine monophosphate residues, a modification important for RNA processing and stability. It shares about 60% amino acid identity with poly(A) polymerase II (PAPII) and localizes exclusively to the nucleus. PAPOLG exhibits both nonspecific and CPSF/AAUAAA-dependent polyadenylation activity[1][2]. Defects or deletions affecting PAPOLG are associated with rare genetic syndromes and acute cystitis, but there are no direct indications of approved or investigational drugs targeting this enzyme, nor clinical biomarkers or specific safety concerns documented to date[1].
