Overview

Poly(A) polymerase non-canonical PAPD5 (PAPD5), also known as TENT4A or TRF4-2, is an enzyme involved in RNA surveillance and quality control. It catalyzes the addition of short oligo(A) or oligo(U) tails to RNA substrates, marking them for degradation by the exosome. It plays a role in histone mRNA turnover and regulates telomerase RNA component (TERC) stability. PAPD5 is a therapeutic target in diseases like dyskeratosis congenita and viral infections, with inhibitors showing promise in restoring TERC levels and reducing viral loads.

Mechanism of action

Inhibition of PAPD5/7 activity, leading to increased TERC levels and improved telomerase function; impaired viral RNA tailing activity, leading to destabilization.
