Poly(ADP-ribose) polymerase 14 (PARP14) is a member of the PARP family of enzymes, primarily catalyzing mono-ADP-ribosylation. It contains RNA recognition motifs, macro domains, a WWE domain, and a catalytic domain. PARP14 participates in DNA damage response, transcriptional regulation (STAT6), immune modulation (Th2 responses), and metabolic reprogramming (Warburg effect). It is implicated in cancer progression/metabolism/resistance and allergic inflammation, making it a potential therapeutic target.
