Poly(rC)-binding protein 4 (PCBP4) is a member of the KH domain (K homology domain) protein subfamily, also referred to as alpha-CPs, which specifically bind to C-rich pyrimidine regions in RNA[1][3][5]. PCBP4 is induced by the p53 tumor suppressor and is primarily localized to the cytoplasm, lacking nuclear localization signals found in related proteins[1][3][5]. The protein plays a crucial role in post-transcriptional RNA regulation, influencing the stability and expression of target mRNAs, including the cyclin-dependent kinase inhibitor p21, by binding to CU-rich sequences within their 3' untranslated regions[2][4]. PCBP4 can suppress cell proliferation by promoting apoptosis and enforcing G2-M cell cycle arrest. Multiple alternatively spliced transcript variants of PCBP4 exist, but only some have been fully characterized. Altered function or expression of PCBP4 has been implicated in several cancers and is a downstream effector of p53-mediated tumor suppression[1][2][3][5].
