Overview

Poly (ADP-ribose) polymerase 1 (PARP1) is a multifunctional nuclear enzyme that plays a central role in DNA repair, chromatin remodeling, transcription regulation, cell cycle control, and programmed cell death. It uses NAD+ to catalyze the transfer of ADP-ribose units onto itself and other target proteins (PARylation). PARP1 acts as a first responder to DNA single-strand breaks (SSBs), binding rapidly to damaged sites and recruiting various DNA repair factors. It is a key target in cancer therapy, particularly in tumors deficient in homologous recombination repair genes like BRCA, where PARP inhibition leads to synthetic lethality.
