PC2 is essential for maintaining the structural integrity and function of renal tubules, primarily through its role in the primary cilia and endoplasmic reticulum (PubMed: 31578150). Mutations in PKD2 mRNA, which account for approximately 15% of Autosomal Dominant Polycystic Kidney Disease (ADPKD) cases, typically lead to a loss of functional PC2 protein, triggering cyst formation and progressive renal failure (NIH: Gene ID 5311). Therapeutic targeting of PKD2 mRNA is an emerging field, primarily utilizing RNA-based strategies to enhance the expression of the functional protein. For example, anti-microRNA oligonucleotides like RGLS8429 target miR-17, a microRNA that binds to and suppresses PKD2 mRNA, thereby increasing its stability and translation (Regulus Therapeutics, 2023). This approach aims to restore polycystin signaling to slow the progression of ADPKD and reduce total kidney volume.
