The Porcine circovirus type 2 (PCV2) capsid protein, encoded by open reading frame 2 (ORF2), is the sole structural component of the viral capsid and the primary target for the host's immune system (UniProt P0C6V4). It self-assembles into icosahedral virus-like particles (VLPs) and mediates viral attachment to host cells by binding to glycosaminoglycans like heparan sulfate (PubMed: 17437658). PCV2 is the causative agent of Porcine Circovirus Associated Diseases (PCVAD), including Postweaning Multisystemic Wasting Syndrome (PMWS), which causes severe economic impact in the global swine industry (NCBI: NBK535444). As the major immunogen, the ORF2 protein contains key neutralizing epitopes, making it the central component of commercial subunit vaccines such as Ingelvac Circoflex and Porcilis PCV (PubMed: 22405466). These vaccines work by eliciting neutralizing antibodies that block viral entry and reduce viral load in infected pigs. Ongoing research also explores the protein as a target for entry inhibitors and novel diagnostic assays to monitor herd health.
