The Potato virus X coat protein (PVX CP) is a 25 kDa structural protein derived from a plant virus that has been repurposed as a potent immunostimulatory carrier for therapeutic vaccines (Lico et al., 2015; PubMed: 25613745). It naturally assembles into filamentous virus-like particles (VLPs), providing a robust scaffold for the multivalent display of fused or conjugated antigens, which significantly enhances their visibility to the immune system (McCormick et al., 2008; PubMed: 18599451). Beyond its role as a physical carrier, PVX CP acts as a molecular adjuvant by triggering innate immune signaling pathways that promote the maturation of dendritic cells and the subsequent activation of robust T-cell and B-cell responses (Savonne et al., 2016; PubMed: 27461114). This dual functionality makes it a valuable component in the development of vaccines for cancer, such as those targeting B-cell malignancies or HER2-positive tumors, as well as for infectious diseases (Uhde-Holzem et al., 2010; PubMed: 20624193). In clinical and preclinical settings, PVX CP-based fusions are designed to overcome immune tolerance to self-antigens or poorly immunogenic viral proteins (ClinicalTrials.gov, NCT00006012).
Acts as a high-density multivalent display platform for antigens and functions as a molecular adjuvant by stimulating innate immune pathways, likely through pattern recognition receptors (PRRs), to enhance T-cell and B-cell responses.
