PANVAC is a therapeutic cancer vaccine platform designed to stimulate the immune system to recognize and destroy cancer cells [1, 3]. It consists of two recombinant poxvirus vectors—vaccinia (PANVAC-V) and fowlpox (PANVAC-F)—administered in a prime-boost regimen [2, 4]. Both vectors carry transgenes for two tumor-associated antigens, Carcinoembryonic antigen (CEA) and Mucin 1 (MUC1), which are frequently overexpressed in various adenocarcinomas [3, 8]. Additionally, the vaccine incorporates a triad of costimulatory molecules (TRICOM) comprising B7.1, ICAM-1, and LFA-3 to enhance the activation of T-lymphocytes [4, 6]. By presenting these antigens in a highly immunogenic context, PANVAC aims to overcome immune tolerance and generate a robust, antigen-specific cytotoxic T-cell response against tumors such as pancreatic, colorectal, and breast cancers [6, 10]. Clinical trials have evaluated its safety and efficacy both as a monotherapy and in combination with other agents like Sargramostim or chemotherapy [2, 12]. While it is often referred to as a target in some contexts, PANVAC is technically a therapeutic agent that targets specific biological molecules [1, 13].
