PRAME425–433 peptide presented by HLA-A*02:01 is a specific peptide-major histocompatibility complex (pMHC) target utilized in advanced cancer immunotherapies. The peptide, with the sequence SLLQHLIGL, is a proteasomal cleavage product of the Preferentially Expressed Antigen in Melanoma (PRAME), a well-characterized cancer-testis antigen [2, 13, 15]. While PRAME is highly overexpressed in a wide range of solid and hematological malignancies—including melanoma, sarcoma, and acute myeloid leukemia—its expression in healthy adult tissues is largely restricted to the testis, making it an ideal target for minimizing off-tumor toxicity [1, 3, 4, 7]. This pMHC complex is specifically recognized by engineered T-cell receptors (TCRs) in therapies such as IMA203 (a TCR-T cell therapy) and bispecific T-cell engagers like IMC-F106C [7, 11, 12]. Upon binding, these therapeutic agents redirect T-cells to recognize and eliminate tumor cells presenting the PRAME425–433 epitope [11, 17]. Clinical evaluations have demonstrated significant anti-tumor activity and durable responses in patients with heavily pretreated metastatic melanoma and other PRAME-positive cancers [7, 12]. However, therapeutic application requires careful monitoring for safety concerns such as cytokine release syndrome (CRS) and potential cross-reactivity with similar peptides in healthy tissues [7, 9, 17].
