Overview

Precursor microRNA-449b (pre-miR-449b) is a non-coding RNA molecule that is enzymatically processed into mature miR-449b, a member of the miR-449 family located within the CDC20B gene on chromosome 5 (Lize et al., 2011, PubMed: 21170278). It functions primarily as a tumor suppressor by post-transcriptionally regulating genes involved in cell cycle progression and apoptosis, such as CDK6, CDC25A, and SIRT1 (Bou Kheir et al., 2011, PubMed: 21571556). In various cancers, including lung and gastric malignancies, miR-449b is frequently downregulated, which contributes to increased cell proliferation and chemoresistance. Additionally, pre-miR-449b plays a vital role in multiciliogenesis in the airway and reproductive tracts by inhibiting the Notch signaling pathway (Marcet et al., 2011, PubMed: 21844874). Therapeutic interventions targeting this molecule typically involve miRNA mimics to restore its suppressive activity in cancer or antagomirs to modulate its function in other contexts. However, the clinical application of miR-449b-based therapies faces significant hurdles, including the need for efficient tissue-specific delivery systems and the mitigation of potential off-target effects on the transcriptome.
