Programmed cell death protein 1 (PD-1) is a type I transmembrane receptor expressed on the surface of T cells and other immune cells in dogs. It acts as an immune checkpoint, suppressing T cell activity by binding to its ligands, PD-L1 and PD-L2, which can be exploited by tumors and pathogens to evade immune attack. Canine PD-1 is structurally and functionally homologous to human PD-1 and is a validated target for immunotherapy in comparative oncology. Monoclonal antibodies and nanobodies have been developed to block the interaction between canine PD-1 and its ligands, showing promise in restoring antitumor immunity in canine cancer models.
