Proinsulin antigen presentation refers to the process by which fragments of the proinsulin protein are processed and displayed on major histocompatibility complex (MHC) molecules by antigen-presenting cells (APCs). This presentation is central to immune recognition in type 1 diabetes (T1D), where loss of tolerance to proinsulin leads to autoimmune destruction of pancreatic β-cells. Tolerance induction strategies aim to present proinsulin in a way that renders autoreactive T cells unresponsive or deletes them, thereby preventing or ameliorating autoimmunity.
Enforced/tolerogenic presentation leads to deletion/anergy/regulatory conversion of autoreactive T cells.
