Prolactin receptor (PRLR) mRNA is the messenger RNA transcript that encodes the prolactin receptor protein, a member of the class I cytokine receptor family. The prolactin receptor is essential for mediating the effects of prolactin, a hormone involved in over 300 biological functions, including lactation, reproduction, and immune modulation (UniProt P16471). In diseases such as endometriosis and breast cancer, the prolactin signaling pathway is often upregulated, contributing to lesion growth and tumor progression (PubMed: 31533971). Targeting the mRNA transcript itself using antisense oligonucleotides (ASOs) allows for the sequence-specific degradation of the message, leading to a significant reduction in the translation of the PRLR protein. This therapeutic strategy, exemplified by candidates like IONIS-PRLR-LRx, aims to inhibit the pathological effects of prolactin signaling while potentially offering a more targeted profile than broad hormonal antagonists (ClinicalTrials.gov: NCT04143399).
