Overview

Proliferation-associated protein 2G4 (PA2G4), also known as ErbB3-binding protein 1 (EBP1), is a multifunctional protein that plays a pivotal role in regulating cell growth, apoptosis, and differentiation [1, 2]. It exists as two distinct isoforms, p48 and p42, which often exhibit antagonistic functions; the p48 isoform typically promotes tumor cell proliferation and survival, whereas the p42 isoform can act as a tumor suppressor [3, 4]. PA2G4 functions as an RNA-binding protein involved in ribosome biogenesis and as a transcriptional co-regulator that interacts with various signaling molecules, including the ErbB3 receptor and androgen receptor [1, 5]. In the context of therapeutic development, PA2G4 mRNA is targeted using RNA-based strategies such as antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) to downregulate the expression of the pro-tumorigenic p48 protein [4, 6]. High levels of PA2G4 expression are frequently observed in various malignancies, including prostate, breast, and hepatocellular carcinomas, and are often correlated with poor clinical outcomes [3, 7]. Consequently, modulating PA2G4 levels or its mRNA stability represents a potential therapeutic avenue for treating advanced cancers and certain neurodegenerative conditions [4, 8].
