Overview

Proline, histidine and glycine-rich protein 1 (PHGR1) is a small, glycosylated protein expressed primarily in mature epithelial cells of the gastrointestinal (GI) tract, especially in the colon and small intestine[1][2][3][5]. PHGR1 localizes to both the cytoplasm and nucleus, with nuclear localization increasing during epithelial cell differentiation[2][5]. Transcriptomic and gene ontology analyses indicate roles in intracellular transport and metabolic processes in GI epithelial cells[1][2]. Knockdown of PHGR1 leads to significant changes in gene expression related to transport, localization, and metabolic functions[2]. PHGR1 is not a receptor, enzyme, transporter, or classical therapeutic target, but it is considered a promising biomarker for detection of lymph node metastases in colorectal cancer and may have utility in cancer diagnostics[1][2]. There is no current evidence of direct drug interactions or known safety concerns described for this protein.
