Overview

Protein kinases are a large family of enzymes that catalyze the transfer of phosphate groups from ATP to specific amino acids (serine, threonine, or tyrosine) on substrate proteins. Isoform-specific protein kinases refer to distinct variants (isoforms) within a kinase family that arise from different genes or alternative splicing and often have unique regulatory properties, subcellular localizations, and physiological functions. Dysregulation/mutation of specific kinase isoforms contributes to diseases such as cancer/metabolic disorders/neurodegeneration. Development of inhibitors/disruptors that target individual kinase isoforms offers potential therapeutic advantages by minimizing off-target effects seen with pan-inhibitors.
