Overview

Protein kinase C (PKC) is a family of serine/threonine kinases that serve as pivotal mediators in platelet signaling pathways (Harper & Poole, 2010, FEBS Letters). In human platelets, multiple isoforms including PKCα, PKCβ, PKCδ, and PKCθ are expressed and activated by secondary messengers like diacylglycerol (DAG) and calcium (Ca2+) (PubChem, CID 5281). These kinases regulate essential platelet functions such as the release of alpha and dense granules, the activation of the integrin αIIbβ3 receptor, and subsequent platelet aggregation (UniProt, P17252). Because of its central role in thrombus stabilization and growth, PKC is a significant therapeutic target for treating cardiovascular disorders and preventing arterial thrombosis (PubMed, 20157909). However, the high degree of homology between PKC isoforms and their widespread expression in other tissues, such as the immune and nervous systems, necessitates the development of isoform-specific inhibitors to minimize adverse effects like bleeding or immunosuppression (StatPearls, Protein Kinase C).
