Overview

Protein kinase C beta (PKCβ) is a member of the classical PKC subfamily of serine/threonine kinases, which are activated by calcium and diacylglycerol (UniProt P05771). It plays a pivotal role in various signal transduction pathways, particularly in B-cell receptor signaling and vascular endothelial growth factor (VEGF) signaling (PubMed: 16557271). In oncology, PKCβ is frequently overexpressed in B-cell malignancies like diffuse large B-cell lymphoma (DLBCL), where it promotes cell survival and proliferation through the activation of the NF-kappaB pathway (PubMed: 15905474). In metabolic diseases, its activation is linked to the microvascular complications of diabetes, such as retinopathy and nephropathy, due to its role in mediating glucose-induced vascular damage (PubMed: 17065637). Therapeutic strategies primarily focus on small-molecule inhibitors to block its catalytic activity, though achieving isoform specificity remains a challenge (PubChem CID: 176167).
