Target intelligence / Profile preview

Protein phosphatase 3 regulatory subunit B, beta (PPP3R2)

Target
PPP3R2
Molecular classification
Enzyme regulator, Regulatory subunit of serine/threonine phosphatase, Calcineurin complex component
01

Overview

Protein phosphatase 3 regulatory subunit B, beta (PPP3R2) is a regulatory subunit of calcineurin (protein phosphatase 3), a calcium- and calmodulin-dependent serine/threonine protein phosphatase that plays a critical role in signal transduction pathways, especially in T-cell activation and neuronal signaling[8][10]. The beta subunit confers calcium sensitivity to the calcineurin enzyme complex and participates in the regulation of immune response and neurological signaling[8]. The gene encoding this subunit is protein coding, with related roles in pathways such as cAMP-Dependent PKA activation and TCR signaling[8][10].

Mechanism of action

Immunosuppressive drugs (e.g., cyclosporin, tacrolimus) inhibit calcineurin phosphatase activity by binding to immunophilins, which then block the calcineurin complex (including the regulatory subunit B, beta), suppressing T cell activation and immune response

03

Biological functions

Calcium-dependent regulation of protein serine/threonine phosphatase activityCalcineurin-mediated signal transductionModulating calcium signaling pathways
04

Disease associations

Neurodegenerative disease (implicated via calcineurin signaling pathway)Immunological regulation (immune response via calcineurin pathway)Other (fertility: penetration of zona pellucida in reproduction)
05

Safety considerations

Long-term inhibition of calcineurin can result in nephrotoxicity, neurotoxicity, and immunosuppression due to disruption of normal calcium signaling and T-cell regulation
06

Interacting drugs

Cyclosporin A (by inhibition of calcineurin, though it binds to the catalytic subunit, the regulatory subunit B is required for full function)

Biomarkers

No direct clinical use as a biomarker, but altered calcineurin composition (including PP3R2) can affect cellular calcium signaling or be studied in the context of immune and neurodegenerative disorders
