Overview

Protein tyrosine kinase 2 beta (PTK2B), also known as Focal adhesion kinase 2 (FAK2) or Proline-rich tyrosine kinase 2 (Pyk2), is a non-receptor tyrosine kinase that plays a pivotal role in integrating signals from the extracellular matrix and soluble factors [UniProt]. Unlike its relative FAK1, PTK2B is primarily activated by increases in intracellular calcium and stress stimuli, making it a key mediator in the central nervous system and immune cells [UniProt, NIH]. It regulates essential cellular processes including adhesion, motility, and survival by acting as both a signaling enzyme and a molecular scaffold [UniProt]. In oncology, PTK2B is frequently overexpressed and contributes to tumor progression, metastasis, and resistance to therapies like tamoxifen [NIH, Dovepress]. Beyond cancer, it is recognized as a genetic risk factor for Alzheimer's disease and a potential target for treating inflammatory conditions and heterotopic ossification [eLife, Frontiers]. Current therapeutic strategies focus on small-molecule inhibitors, many of which are dual FAK1/FAK2 inhibitors, to disrupt these pathological signaling pathways [ResearchGate, Patsnap].
