Overview

CD45RC is one of several isoforms of the leukocyte common antigen (CD45) produced by alternative splicing that includes exon C of the gene. CD45 proteins are type I transmembrane glycoproteins and are among the most abundant protein markers of lymphocytes (comprising up to 10% of surface protein). All CD45 isoforms share a large, highly conserved cytoplasmic domain (~700 amino acids) harboring dual protein tyrosine phosphatase activities, essential for dephosphorylation events controlling immune cell activation and differentiation. The extracellular domain differs between isoforms depending on splice variants, impacting function and antibody recognition. CD45RC plays a role in regulating immune signaling, notably in T and B cell activation and differentiation. Its expression profile can facilitate discrimination of lymphocyte subsets and serves both as a diagnostic marker (immunophenotyping) and as a putative therapeutic target in diseases where immune modulation is required, including specific cancers, inflammatory disorders, and transplantation rejection.

Drugs and antibodies interacting with CD45RC may act by modulating its tyrosine phosphatase activity, altering immune cell signal transduction, or targeting immune cells for depletion (antibody-dependent cytotoxicity). Immunomodulation by selective targeting of immune cell subsets based on isoform expression.
