Overview

The Protein-tyrosine phosphatase 1B (PTP1B) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the PTPN1 transcript that has been targeted for the treatment of metabolic disorders. PTP1B is a non-receptor phosphotyrosine phosphatase that acts as a major negative regulator of insulin and leptin signaling by dephosphorylating the insulin receptor and Janus kinase 2 (JAK2). The 3'-UTR of the PTP1B mRNA contains specific sequences that influence transcript stability and translation, making it an ideal site for antisense oligonucleotide (ASO) intervention. Drugs like ISIS 113715 (IONIS-PTP1B_Rx) are designed to hybridize with this region, triggering RNase H-mediated degradation of the mRNA and subsequently reducing the cellular levels of the PTP1B protein. This reduction enhances the sensitivity of insulin and leptin pathways, leading to improved glucose control and weight loss in patients with type 2 diabetes and obesity. Although 'PTB1' is a common synonym for the RNA-binding protein PTBP1, in the context of therapeutic 3'-UTR targeting, it is frequently used as a shorthand or misspelling for PTP1B.
