Protocadherin gamma subfamily A member 4 (PCDHGA4) is a membrane protein of the calcium-dependent cell adhesion family, specifically part of the protocadherin gamma cluster on chromosome 5. It mediates neural cell-cell adhesion and is thought to play a critical role in the establishment and maintenance of specific neuronal connections in the brain. Each protocadherin gamma subfamily gene encodes extracellular cadherin domains and a shared cytoplasmic region. These proteins likely contribute to neuronal cell surface diversity, possibly influencing synaptic specificity, self-recognition, and single-neuron identity. Although mutations in the protocadherin gene family have been linked to a variety of neurodevelopmental disorders, direct evidence for PCDHGA4 as a causative gene is limited. Expression is highest in brain tissue—including regions such as hippocampus, amygdala, and cortex.
