Overview

Protoporphyrin IX (PpIX) cellular targets refer to the collective subcellular structures damaged during photodynamic therapy (PDT) when PpIX is activated by light. PpIX is a potent endogenous photosensitizer that accumulates preferentially in neoplastic cells following the administration of precursors like 5-aminolevulinic acid (5-ALA) [Source: National Cancer Institute]. Upon exposure to specific wavelengths of light, PpIX transfers energy to molecular oxygen, generating highly reactive singlet oxygen [Source: PMC3076234]. Because singlet oxygen has an extremely short diffusion radius (approximately 20 nm), the primary targets are the organelles where PpIX is localized, specifically the mitochondria, endoplasmic reticulum, and lysosomes [Source: PubMed 11460120]. The resulting oxidative damage to lipids and proteins within these components triggers irreversible cell death via apoptosis, necrosis, or autophagy-associated pathways [Source: PMC4090446].
