Protoporphyrin IX (PpIX)-sensitized cellular components refer to the specific intracellular structures, such as mitochondrial membranes, the endoplasmic reticulum, and the plasma membrane, that become susceptible to oxidative damage during photodynamic therapy (PDT) [StatPearls, 2023]. PpIX is a naturally occurring porphyrin and a potent photosensitizer that accumulates preferentially in malignant or rapidly dividing cells following the administration of precursors like aminolevulinic acid (ALA) [PubChem, 2024]. Upon irradiation with specific wavelengths of light, the accumulated PpIX molecules transfer energy to molecular oxygen, generating highly reactive singlet oxygen and other reactive oxygen species (ROS) [National Cancer Institute, 2021]. These ROS cause immediate, localized damage to the sensitized cellular components, leading to cell death through apoptotic or necrotic pathways [Journal of Clinical Medicine, 2020]. This targeted approach is widely used in the treatment of dermatological conditions like actinic keratosis and various cancers, including basal cell carcinoma and bladder cancer [StatPearls, 2023]. The therapeutic specificity is achieved through both the selective metabolic accumulation of PpIX in target tissues and the precise application of light to the treatment site [National Cancer Institute, 2021].
